چکیده

Cotton is a cash crop of many countries as it serves source fiber, edible oil, and seedcake. It grown in diverse range environment. Bacillus thuringiensis (Bt) gram positive bacterium found different habitats but mostly soil. This produces endotoxin which harmful for various insects. Numerous crystal-forming strains Bt exist nature, not all them are efficient against insect pests. cotton prevalent internationally because less attack. The key objective this research was to analyze isolates residing soil fields Multan Bahawalpur districts identify on the basis morphology. To attain this, samples were collected from (Gup wala, Mosey Wala, Taloki Hafiz Choudary Basti Mangla Mari, Sharkha Lawain Ghulamo Wala) district (Basti Deewan Wali, Check No 8, Tariqabad, Khaji Khandin, Rammo Sultanpur, Khurampur, Jahanpur). About 178 colonies bacteria eighteen appeared Nutrient Agar plates. screening based morphology yielded 18 Bt. We performed endo-toxin production experiment confirmed presence parasporal crystals identified B. colonies. Colony forming units represented statistically significant results. Based morphology, eight out Results current study revealed that isolates, 8 bacterial media had an increased elevation, round, white, slimy smooth edges. Toxicity test resistance leaf worm second instar larvae two (BtS2, BtS7) toxic showed 39.25% mortality. will help local can be used indigenous pest control.

Isolation, characterization and molecular identification of Bacillus thuringiensis Alex-13 isolated from Egypt against Spodoptera littoralis

The bacterium Bacillus thuringiensis widely used as bio-pesticide. The specific toxic activity to insects and another microbe(s) regarding the existing of crystals that have different morphologies, sizes, numbers and compositions, as Bt strains the crystals have more different proteins, which contain encoded Cry distinct genes. In this work we isolated 334 colonies from 59 soil samples in 13 lo...

Silencing of midgut aminopeptidase N of Spodoptera litura by double-stranded RNA establishes its role as Bacillus thuringiensis toxin receptor.

Insecticidal crystal proteins of Bacillus thuringiensis bind to receptors in the midgut of susceptible insects leading to pore formation and death of the insect. The identity of the receptor is not clearly established. Recently a direct interaction between a cloned and heterologously expressed aminopeptidase (slapn) from Spodoptera litura and the Cry1C protein was demonstrated by immunofluoresc...

Effectiveness of Dead-Spore Bacillus thuringiensis Formulation Against Diamondback Moth

We developed a process of producing a dead-spore Bacillus thuringiensis Berliner var. kurstaki HD-1 formulation, Toarow CT, and studied several aspects t o improve the productivity of crystal toxin and the insecticidal potency after sterilization to kill spores. The production of cell mass or crystal toxin was highly influenced by aeration and agitation of the culture. The culture broth, within...

Synergistic activity of a Bacillus thuringiensis delta-endotoxin and a bacterial endochitinase against Spodoptera littoralis larvae.

In an attempt to increase the insecticidal effect of the delta-endotoxin crystal protein CryIC on the relatively Cry-insensitive larvae of Spodoptera littoralis, a combination of CryIC and endochitinase was used. CryIC comprising the first 756 amino acids from Bacillus thuringiensis K26-21 and endochitinase ChiAII encoded by Serratia marcescens were separately produced in Escherichia coli carry...

Interaction between Calcium Ions and Bacillus thuringiensis Toxin Activity against Sf9 Cells (Spodoptera frugiperda, Lepidoptera).

The effects of calcium ions and modulators of calcium movement on Bacillus thuringiensis insecticidal protein toxicity were investigated with Sf9 cells (Spodoptera frugiperda, fall armyworm) by a new B. thuringiensis toxicity assay based on measurement of fluorescence of ethidium homodimer, a high-affinity DNA stain. CryIC toxicity was substantially stimulated by extracellular calcium in a dose...
